Abstract
In the first part of this talk I will introduce the development of a new robust image-based adaptive optics (AO) two-photo fluorescence microscope. By modulating the intensity or phase of light rays at multiple pupil segments in parallel to determine their phase gradients, we were able to measure the aberration in the biology organism and applied it to fluorescent-protein-labeled structures of arbitrary complexity. This method allows us to obtain diffraction-limited resolution in various samples in vivo, from c. Elegant, zebrafish to the functional imaging of mouse visual cortex. For the strongly scattering mouse brain, a single aberration correction improves structural and functional imaging of fine neuronal processes over a large imaging volume.
In the second part, I will describe the development of a microscopy framework that integrates high-speed, multi-view light-sheet imaging of the entire nervous system with concurrent cellular-resolution optical manipulation using wide-field patterned illumination and three-dimensional multi-photon excitation. Furthermore, by developing acomputational control layer that analyzes and classifies calcium imaging data in real time, we designed experiments that facilitate real-time image-based behavior detection and closed-loop optogenetic manipulation of neuronal activity during behavior.
